Overview

Pericentrin (PCNT) is a large, conserved scaffold protein of the pericentriolar material (PCM) at the centrosome, essential for organizing centrosomal matrix components during mitosis[1][3][4][5][6]. It serves as an anchor for a wide number of regulatory and structural proteins, including the γ-tubulin ring complex, facilitating microtubule nucleation and proper mitotic spindle formation for cell division[1][3][4][5][6]. PCNT is also implicated in DNA damage response via its interactions at the centrosome. Mutations in the PCNT gene are responsible for several congenital disorders featuring growth defects, such as microcephalic osteodysplastic primordial dwarfism type II and Seckel syndrome 4[2][3][4][6]. Increased PCNT expression has been observed in certain cancers, where it is associated with centrosomal abnormalities and chromosomal instability[2][6].
