Overview

Pericentriolar material 1 protein (PCM1) is a 228 kDa scaffolding protein fundamental for the structure and function of centriolar satellites, which are small cytoplasmic granules involved in anchoring microtubules to the centrosome and recruiting key regulatory proteins. PCM1 is crucial for proper cell division, cilium assembly, and organization of the microtubule network. It acts as a hub for spatial recruitment of centrosomal proteins—such as centrin, pericentrin, and ninein—and for protein complexes critical to cell cycle progression and cilia formation. Genetic mutations in PCM1 are associated with increased risk of certain cancers and schizophrenia, and knockout models show a requirement for PCM1 in male fertility and ciliary motility. PCM1 is regulated by phosphorylation, polyubiquitination, and other post-translational modifications, which adjust its structural and functional contributions to centrosome biology. PCM1 is not targeted by current drugs and is used primarily in research as a marker and functional regulator of centriolar satellites and centrosome integrity.
