Periodic tryptophan protein 2 homolog (PWP2) is a conserved, nucleolar protein that is an essential component of the small subunit (SSU) processome, the primary precursor of the small eukaryotic ribosomal subunit. PWP2 functions within the UtpB subcomplex and plays a central role in the biogenesis and assembly of the 18S rRNA-containing small ribosomal subunit by facilitating rRNA folding, modification, and processing events. It is classified as a WD-repeat-containing protein, which enables interactions with RNA and numerous protein partners within the nucleolus. Genetic disruption of PWP2 affects ribosome assembly and leads to cellular and developmental defects. No current drugs are known to target PWP2, and it is not considered a therapeutic target as of 2025.
