Overview

The periodontal ligament (PDL) is a highly specialized, vascularized, cellular connective tissue located between the cementum covering the tooth root and the alveolar bone of the jaw[1][3][7]. Comprised primarily of collagen fiber bundles (mainly type I, III, and V collagens) and interfiber ground substance, it serves to attach and support the root of each tooth, acting as a shock absorber during chewing, distributing mechanical stresses, and providing nutrients via its extensive vascular and lymphatic network[1][4][5][7]. Fibroblasts are the predominant cells, but defense cells (macrophages, neutrophils, lymphocytes), cementoblasts, osteoblasts, and epithelial rest cells are also present[1][4]. The PDL is essential for normal tooth function, tooth movement (such as in orthodontics), bone remodeling, and sensory feedback during mastication[5][7]. Degradation or dysfunction leads to periodontal disease and loss of tooth attachment[3][5][7]. In summary, Periodontal ligament is not a molecular therapeutic target but rather a complex tissue—multiple proteins and cell types within it may serve as research or therapeutic targets in the context of dental medicine[3][7].
