Overview

Peripheral blood mononuclear cells (PBMCs) are a collection of blood cells with single round nuclei, including lymphocytes (T cells, B cells, and natural killer cells), monocytes, and a minority of dendritic cells, making up the key cellular components of the immune system involved in adaptive and innate responses. PBMCs are isolated from blood, typically using density-gradient centrifugation (e.g., Ficoll-Paque), and are widely used in immunological research to study immune cell interactions, disease mechanisms, drug responses, and vaccine efficacy. PBMCs are not a molecular drug target, but an analytical and experimental model in immunology, cancer research, infection monitoring, and autoimmune diseases

Biological functions

Immune response (both innate and adaptive immunity)Pathogen recognitionAntigen presentationSignal transduction (via constituent cell types)Cell-mediated cytotoxicityAntibody production (by B cells)Phagocytosis (by monocytes/macrophages)
03

Disease associations

Cancer (immunotherapy research, immune profiling)Infection (HIV, viral markers, screening, etc.)Inflammation (autoimmune diseases, study of inflammatory responses)Other (immune monitoring in transplantation, vaccine response)
04

Safety considerations

PBMCs themselves do not raise direct safety concerns, but their use in research may involve biohazard safety associated with blood samples (infection transmission risks, contaminated samples)
05

Biomarkers

Specific immune cell markers (CD3 for T cells, CD19 for B cells, CD14 for monocytes, CD56 for NK cells, etc.)Cytokine profiles (IL-2, IFN-gamma, TNF-alpha, etc. as functional readouts)
