Overview

The Peripheral blood mononuclear cell (PBMC) IL-4-induced IgE synthesis pathway describes the cellular and molecular mechanisms by which interleukin-4 (IL-4) stimulates PBMCs, specifically B cells, to produce immunoglobulin E (IgE). IL-4 binds to its receptor on B cells, activating STAT6 and other downstream signaling molecules, leading to class switch recombination to IgE. This process is enhanced by CD40L signaling from T cells and modulated by other cytokines. The pathway is crucial in allergic responses, and its dysregulation contributes to atopic diseases.

Mechanism of action

Varies depending on the specific drug. Anti-IL-4 antibodies neutralize IL-4, preventing its binding to the IL-4 receptor. IFN-gamma and IFN-alpha inhibit IgE synthesis through various mechanisms. Prostaglandin E2 and retinoic acid also suppress the pathway.
