Overview

Peripheral receptors on human nasal chemosensory neurons are a specialized group of sensory receptors located in the nasal chemosensory epithelium, primarily within the nasal passages. These receptors serve as the primary therapeutic target for a novel class of intranasal compounds known as pherines, which include fasedienol and itruvone. Unlike traditional neuropsychiatric medications that require systemic absorption and penetration of the blood-brain barrier, these receptors facilitate a nose-to-brain mechanism of action. Upon activation by an agonist, they trigger rapid neural impulses through the olfactory bulb to the limbic system, specifically targeting the amygdala and hypothalamus to modulate circuits involved in fear, anxiety, and autonomic responses. This localized interaction allows for rapid therapeutic onset, often within minutes, while avoiding the systemic side effects and abuse potential associated with traditional CNS drugs. These receptors are currently being investigated for the treatment of social anxiety disorder, major depressive disorder, and vasomotor symptoms associated with menopause.
