Peroxidasin-like protein (PXDNL), also known as Vascular Peroxidase 2 (VPO2), is a heme-containing protein that is predominantly expressed in the human heart (UniProt: A6NHR9). Although it shares high sequence homology with Peroxidasin (PXDN), PXDNL lacks the essential distal histidine residue required for the standard peroxidase catalytic cycle, leading to its classification as a catalytically inactive pseudoperoxidase (PubMed: 24510963). It is localized to the extracellular matrix (ECM) of cardiomyocytes where it is thought to play a structural or regulatory role rather than an enzymatic one (PubMed: 21464248). Research suggests that PXDNL may act as a dominant-negative regulator of PXDN, thereby modulating the formation of sulfilimine bonds in collagen IV and influencing the stability of the cardiac ECM (PubMed: 28611103). Dysregulation of PXDNL has been associated with various cardiovascular pathologies, including dilated cardiomyopathy and heart failure, suggesting it may be a relevant target for managing cardiac remodeling (PubMed: 30265412). Currently, there are no approved drugs or clinical-stage compounds that specifically target this molecule.
