Peroxiredoxin 3 is an essential mitochondrial antioxidant enzyme involved not only in detoxifying ROS but also regulating cell fate decisions under both physiological conditions and disease states such as cancer, metabolic syndrome, inflammation, and drug-induced organ injury.
The active site cysteine reacts with hydrogen peroxide/peroxide substrates forming sulfenic acid. This intermediate forms an intermolecular disulfide bond with another conserved cysteine on an adjacent subunit. Thioredoxin reduces this disulfide bond back to free thiols, completing the catalytic cycle
