Overview

Peroxisomal coenzyme A diphosphatase NUDT7 (NUDT7) is a peroxisomal enzyme and member of the Nudix hydrolase family, characterized by the conserved NUDIX box motif. Its main biochemical function is to hydrolyze coenzyme A (CoA), medium-chain acyl-CoA esters, and oxidized CoA to detoxify potentially harmful nucleotide metabolites and regulate the levels of CoA and acyl-CoA in peroxisomes[2][3][5][6]. NUDT7 is highly expressed in the liver, with lower expression in other tissues such as lung and kidney[2][5]. It may protect cells from oxidative stress by clearing oxidized CoA derivatives, thus maintaining metabolic and redox homeostasis[3][5]. Dysfunction or altered expression of NUDT7 has been associated with neurodegenerative disorders and may be implicated in metabolic stress and cancer biology through redundancy among NUDIX family enzymes[3][7]. First-in-class small molecule inhibitors of NUDT7 have been identified as research tools to probe its function, but there are currently no therapeutic drugs targeting this enzyme clinically[5].
