Overview

Peroxisomal membrane protein 11B (PEX11B) is a transmembrane protein encoded by the PEX11B gene in humans and is essential for peroxisome proliferation and division. It remodels and elongates peroxisome membranes by interacting with specific membrane lipids through its N-terminal amphipathic helices, then assembles and activates multiple division factors, specifically FIS1, MFF, and DRP1. PEX11B can promote peroxisome division even in the absence of MFF, functioning in a FIS1- and DRP1-dependent manner, and can also impact mitochondrial dynamics if targeted to mitochondria. Mutations lead to rare, inherited peroxisomal biogenesis disorders with neurological and developmental phenotypes. PEX11B is found in the peroxisomal membrane and interacts with proteins such as PEX19 for membrane insertion

Mechanism of action

Not applicable. No direct drug mechanism; however, research suggests upregulating PEX11B or MFF might restore peroxisome dynamics in disease, a concept still in preclinical investigation

Biological functions

Peroxisome proliferationPeroxisome membrane elongation and divisionOrganelle dynamics (peroxisome and potential mitochondrial membrane remodeling)Interaction with fission factors (FIS1, MFF, DRP1) for organelle division
04

Disease associations

Other (Peroxisomal biogenesis disorders, notably Zellweger spectrum disorders, have been linked to PEX11B deficiency/disruption)Neurological and developmental symptoms in affected individuals
05

Safety considerations

Therapeutic challenges might include difficulty in pharmacologically modulating peroxisomal biogenesis factors. No direct drug safety concerns are described, as it is not a direct drug target.
06

Biomarkers

Mutations in PEX11B may serve as genetic biomarkers for diagnosis or patient selection in rare peroxisomal biogenesis disorders
