Peroxisomal membrane protein 4 (PXMP4) is a ubiquitously expressed, integral peroxisomal membrane protein of approximately 24 kDa, encoded by the PXMP4 gene in humans. PXMP4 is transcriptionally regulated by peroxisome proliferator-activated receptor alpha (PPARα) and may function in peroxisomal ether lipid metabolism, specifically influencing hepatic levels of alkyldiacylglycerol ether lipids. Knockout studies in mice suggest it is not essential for core peroxisomal β-oxidation activity or bile acid synthesis, but there may be a role in ether lipid and phytanic acid metabolism. Although mutations and hypermethylation of PXMP4 have been observed in several cancers, its precise physiological function and relevance as a disease biomarker or therapeutic target remain largely undefined.
