Peroxisome proliferator–activated receptor gamma coactivator 1-alpha (PGC-1α) is a transcriptional coactivator encoded by the PPARGC1A gene. It is a master regulator of mitochondrial biogenesis and plays a central role in regulating genes involved in energy metabolism. It links external physiological stimuli to cellular adaptation through increased mitochondrial content and activity. Altered function or expression of PGC‑1α has been implicated in several diseases, making it a potential drug target.
PGC‑1α interacts with various transcription factors to enhance their activity on specific gene promoters, leading to coordinated upregulation of genes required for oxidative phosphorylation, fatty acid oxidation, gluconeogenesis, adaptive thermogenesis, cholesterol homeostasis regulation, and blood pressure control mechanisms.
