Overview

Peroxisome proliferator-activated receptor gamma coactivator 1-alpha isoform 4 (PGC-1α4) is a specific protein variant derived from the PPARGC1A gene through alternative splicing and promoter usage (Ruas et al., 2012, Cell). Unlike the well-known PGC-1α1 isoform that regulates mitochondrial biogenesis and oxidative metabolism, PGC-1α4 is uniquely induced by resistance exercise and specifically promotes skeletal muscle hypertrophy (Ruas et al., 2012, Cell). It functions as a transcriptional coactivator that increases the expression of Insulin-like Growth Factor 1 (IGF-1) while simultaneously repressing Myostatin, a key inhibitor of muscle growth (Ydfors et al., 2013, Physiological Reports). This dual action makes it a highly attractive therapeutic target for treating muscle-wasting conditions such as sarcopenia, cancer-associated cachexia, and muscular dystrophies (Roberts et al., 2013, Journal of Applied Physiology). Although no specific pharmacological activators are currently approved for clinical use, PGC-1α4 remains a primary focus for the development of exercise mimetics and anabolic therapies aimed at preserving muscle mass (Boström et al., 2012, Nature).
