Overview

Peroxisome proliferator activity in the liver primarily refers to the function of peroxisome proliferator-activated receptors (PPARs), a family of nuclear receptor proteins that act as transcription factors regulating gene expression in response to ligands such as fatty acids and their derivatives. The PPAR family consists of three main subtypes: PPARα, PPARβ/δ, and PPARγ. These receptors play crucial roles in lipid and glucose metabolism, inflammation, cell proliferation, and differentiation within the liver. Alterations in hepatic PPAR activity are implicated in several diseases, including NAFLD, metabolic syndrome, and diabetes. Activation or inhibition of hepatic PPARs is a therapeutic strategy for metabolic diseases; however, exogenous activation can have side effects limiting clinical use.
