Overview

Pescadillo ribosomal biogenesis factor 1 (PES1) is a nucleolar protein essential for ribosome biogenesis, as part of the PeBoW complex (with BOP1 and WDR12), required for processing precursor rRNA and maturation of the 60S ribosomal subunit. It also regulates cell cycle progression, DNA replication, telomerase assembly, and cellular senescence. PES1 is overexpressed in various cancers and promotes oncogenic growth by modulating cell cycle regulators and estrogen receptor signaling, and by enhancing cancer stem cell characteristics. Its depletion impairs ribosome production and increases sensitivity to chemotherapy. PES1 is a promising molecular target but presents safety concerns due to its central role in normal cellular proliferation.
