Overview

PEST-containing nuclear protein (PCNP) is a short-lived, small nuclear protein (178 amino acids) characterized by the presence of two PEST sequence motifs rich in proline, glutamic acid, serine, and threonine. It plays a critical role in the regulation of the cell cycle, apoptosis, and tumorigenesis across several cancer types by participating in the ubiquitin-proteasome degradation pathway and interacting with regulatory proteins like p53 and NIRF. PCNP modulates key cancer-related pathways, including MAPK, PI3K/AKT/mTOR, and Wnt/β-catenin, and is involved in epigenetic silencing through interactions with histone deacetylases and methyltransferases. Due to its central role in cancer cell proliferation and apoptosis, PCNP is considered a promising, though not yet drugged, target for cancer diagnostics and therapeutics; targeting its stability, ubiquitination, and interactions may represent a novel class of anti-cancer strategies.
