Overview

Peter pan homolog (PPAN) is an evolutionarily conserved protein in humans encoded by the PPAN gene, with homology to yeast SSF1 and the Drosophila Peter Pan gene[1][3][10]. PPAN is essential for cell growth and proliferation and participates in ribosome biogenesis as a Brix domain-containing protein. It is involved in mRNA splicing (especially the second step), mitochondrial homeostasis (through modulation of autophagy), and regulation of apoptosis[1]. Depletion of PPAN leads to increased apoptosis, cell cycle arrest, and upregulation of p53 and its targets. The gene has ubiquitous expression, and its exogenous expression can reduce the anchorage-independent growth of certain tumor cells. Disease association has been noted with narcolepsy and cancer, reflecting its importance in cellular proliferation and differentiation[7][1][10]. There are currently no known drugs, biomarkers, or direct safety considerations linked to therapeutic targeting of PPAN.
