PFKFB3 antisense RNA 1 (PFKFB3-AS1) is a long non-coding RNA located antisense to the PFKFB3 gene. It is overexpressed in cancers such as gastric cancer and correlates with advanced tumor stage and poor prognosis. PFKFB3-AS1 regulates cellular metabolism and proliferation through enhancement of PFKFB3 mRNA and protein expression—PFKFB3 encodes an essential enzyme in the glycolytic pathway that facilitates the Warburg effect in cancer cells. Knockdown of PFKFB3-AS1 reduces PFKFB3 expression, glycolytic flux, and cell proliferation, while its overexpression has the reverse effect. PFKFB3-AS1 is not currently considered a direct therapeutic target, but may serve as a biomarker for cancer progression and prognosis.
Not applicable for drugs targeting PFKFB3-AS1, as none are reported. PFKFB3-AS1 acts by modulating PFKFB3 expression at the RNA level, thereby influencing glycolytic activity and cancer cell proliferation.
