PGM5P3 antisense RNA 1 (PGM5P3-AS1) is a human long non-coding RNA (lncRNA), classified as an antisense RNA that does not encode a protein product[3]. It has been implicated in the regulation of cancer biology, particularly in triple-negative breast cancer (TNBC), where it acts as a tumor suppressor. PGM5P3-AS1 is significantly downregulated in TNBC cells, and experimental studies demonstrate that its overexpression suppresses cell proliferation and migration while promoting ferroptosis, a form of programmed cell death dependent on iron[1][5]. The mechanism involves recruitment of the RNA-binding protein NOP58 to stabilize MAP1LC3C mRNA, thereby promoting ferroptosis and inhibiting malignant progression[1]. In addition, PGM5P3-AS1 has been identified as differentially expressed in gallbladder cancer and may participate in broader developmental and regulatory pathways associated with oncogenesis, although experimental validation in non-breast tumors is limited to computational predictions[2][4]. PGM5P3-AS1 is also annotated by the gene alias FAM233B, but it is not a receptor, enzyme, or canonical therapeutic target molecule. There are currently no known direct drug interactions or established mechanisms of drug action targeting this RNA.
