Overview

PH domain containing endocytic trafficking adaptor 1 (PHETA1) is a protein coding gene whose product localizes primarily to endosomes and is essential for the process of endocytic trafficking, where it regulates the recycling of receptors from endosomes to both the plasma membrane and the trans-Golgi network. PHETA1 interacts directly with the inositol polyphosphate 5-phosphatase OCRL-1 and is involved in endosome organization, receptor recycling, and retrograde endosomal transport. The protein contains a PH (pleckstrin homology) domain, mediating specific protein and lipid interactions related to its trafficking adapter function. Disease associations include Lowe oculocerebrorenal syndrome and Leopard syndrome 1, but PHETA1 itself is not currently considered a direct therapeutic target, nor are there drugs or established biomarkers directly linked to it.
