Overview

The pharyngeal and laryngeal mucosa is the specialized mucous membrane lining the upper aerodigestive tract, encompassing the pharynx and the larynx. It is composed of an epithelial layer—varying from stratified squamous epithelium in high-stress areas to ciliated pseudostratified columnar epithelium in the respiratory portions—supported by a lamina propria rich in blood vessels and lymphoid tissue (StatPearls, 2023). This tissue serves as a primary physiological barrier against inhaled and ingested pathogens, chemical irritants, and mechanical trauma, while also playing a vital role in humidifying air and lubricating the vocal folds for phonation (NIH, 2022). In clinical pharmacology, the mucosa is not a single molecular target but rather a complex environment containing numerous therapeutic targets, such as histamine receptors, sodium channels, and various cytokine receptors. Pathological conditions affecting this region include inflammatory diseases like laryngitis, gastroesophageal reflux-related damage, and malignancies such as squamous cell carcinoma, which often require targeted molecular therapies or topical interventions (PubMed, 2021).

Mechanism of action

Pharmacological agents do not target the mucosa as a single molecule; instead, they interact with specific molecular targets within the tissue. Local anesthetics inhibit voltage-gated sodium channels in mucosal nerve endings; corticosteroids bind to glucocorticoid receptors to reduce inflammation; and mucosal protectants like sucralfate provide a physical barrier over damaged areas (StatPearls, 2023).
