Overview

PHD finger protein 10 (PHF10) is a subunit of the polybromo-BAF (PBAF) subclass of the mammalian SWI/SNF chromatin remodeling complex, notable for harboring a distinctive C-terminal tandem PHD finger domain and an N-terminal SAY domain[1][2]. PHF10 exists as evolutionarily conserved isoforms, one with C-terminal PHD fingers and another with a phosphorylation-dependent SUMO 1 conjugation motif, and each isoform confers tissue-specific functional diversity to PBAF complexes in transcriptional regulation[2]. PHF10 is required for tissue-specific chromatin accessibility, neural progenitor differentiation, osteogenic and odontogenic maturation, and cell proliferation[1][2][3]. Depletion or genetic deficiency of PHF10 leads to impaired differentiation in neural, hematopoietic, and mineralizing tissues (bone, tooth)[1][2]. It is regarded as a critical epigenetic regulator, and its dysfunction is implicated in oncogenesis and developmental disorders through effects on gene expression and cellular differentiation[1][2][3].
