PHD finger protein 12 is a protein-coding gene product characterized by a PHD (plant homeodomain) zinc finger domain. It acts as a transcriptional repressor within SIN3B-type and other chromatin-modifying complexes, stabilizing protein-protein interaction and reading methylation marks on histone H3, notably H3K27ac. PHF12 plays a central role in the negative regulation of gene expression by modulating chromatin state and counteracting the activity of histone acetyltransferases. It operates mainly in the nucleoplasm, recruiting chromatin remodeling complexes and influencing RNA polymerase II transcriptional activity[1][3][5]. Dysregulation of PHF12 or related chromatin remodelers is associated with disease states driven by aberrant gene expression and epigenetic changes.
Drugs targeting similar proteins generally modulate chromatin state by inhibiting or promoting histone acetylation/deacetylation, but no PHF12-targeted agents are listed
