Overview

PHKA1 antisense RNA 1 (PHKA1-AS1) is a long noncoding RNA (lncRNA) arising as a natural antisense transcript to the *PHKA1* gene. It consists of 599 base pairs and lacks protein-coding potential. PHKA1-AS1 is upregulated in non-small cell lung cancer (NSCLC), where increased N6-methyladenosine (m6A) modification stabilizes it. In cancer, PHKA1-AS1 promotes cell proliferation, migration, and metastasis by binding to actinin alpha 4 (ACTN4)—an oncogenic protein—and suppressing its ubiquitin-mediated degradation, thus enhancing ACTN4 protein stability. Inhibition of PHKA1-AS1 reverses these pro-tumorigenic effects. PHKA1-AS1 is studied as a potential biomarker of tumor progression, particularly in NSCLC, but is not itself a therapeutic target, receptor, enzyme, or transporter[1][5][6].
