Overview

Phosducin-like protein 2 (PDCL2) is a conserved eukaryotic cofactor and regulator of the cytosolic chaperonin-containing TCP1 complex (CCT/TRiC). It physically interacts with CCT and modulates its actin, tubulin, and WD-repeat protein folding activity, thereby influencing cytoskeletal assembly and cell cycle progression[1][2]. In mammals, PDCL2 is predominantly expressed in germ cells during spermatogenesis and is required for male fertility; genetic loss in mice leads to infertility due to defective spermiogenesis[2]. Unlike other phosducin-family proteins, PDCL2 does not appear to be critical in G-protein signaling. There are no current therapeutic applications, drug interactions, or established biomarker roles for PDCL2. Its primary biological significance lies in its role as a chaperonin cofactor required for the folding and biogenesis of several essential cellular proteins.
