Overview

Phosphatase and actin regulator 3 (PHACTR3) is a regulatory subunit for protein phosphatase 1 (PP1) that binds both actin and the catalytic subunit of PP1, inhibiting PP1 activity[1][2][3]. PHACTR3 is associated with the nuclear scaffold in proliferating cells and is particularly enriched in neurons, where it regulates cytoskeletal dynamics, dendritic morphology, and axon elongation through independent actin-binding and PP1-binding domains[3][5]. The protein promotes cell spreading and motility by modulating actin polymerization and is involved in neural as well as undifferentiated or transformed cell function[3]. Alterations of PHACTR3, including hypermethylation and deregulated expression, have been implicated in the development of cancers such as colorectal and non-small cell lung cancer, suggesting a role in tumorigenesis and potential utility as a biomarker for early cancer detection[3][5]. No direct therapeutic drugs or known pharmacological inhibitors target PHACTR3 as of now.
