Overview

Phosphatase and tensin homolog (PTEN) mRNA is the messenger RNA transcript responsible for the synthesis of the PTEN protein, a critical tumor suppressor and dual-specificity phosphatase (UniProt P60484). The specific 19–21 nucleotide sequence within this mRNA serves as a target for RNA interference (RNAi) technologies, such as short hairpin RNAs (shRNAs) or small interfering RNAs (siRNAs). When a complementary shRNA guide strand binds to this sequence, it facilitates the recruitment of the RNA-induced silencing complex (RISC), leading to the site-specific cleavage and subsequent degradation of the PTEN mRNA (NCBI Gene 5728). This process effectively silences PTEN expression, resulting in the hyperactivation of the PI3K/AKT/mTOR signaling pathway. While PTEN deficiency is a hallmark of various cancers, targeted knockdown of PTEN mRNA is a significant tool in biomedical research and is being explored for therapeutic applications, such as enhancing axonal regrowth following spinal cord or optic nerve injuries (PubMed 18927399). However, the use of PTEN-targeting agents carries significant safety risks, primarily the potential for inducing uncontrolled cell proliferation and tumorigenesis.
