Overview

Lipin-1 (LPIN1) is a bifunctional protein that plays a pivotal role in cellular lipid metabolism and signaling (UniProt Q14693). It functions as a magnesium-dependent phosphatidate phosphatase (PAP) enzyme, catalyzing the conversion of phosphatidic acid to diacylglycerol, which is a critical step in the biosynthesis of triglycerides and phospholipids (NCBI Gene 10225). Additionally, Lipin-1 acts as a transcriptional coactivator in the nucleus, where it interacts with PPAR-alpha and PGC-1alpha to regulate the expression of genes involved in fatty acid oxidation (PMID: 16432250). Mutations in the LPIN1 gene are the most common cause of severe, recurrent rhabdomyolysis in children, highlighting its essential role in muscle membrane integrity and energy homeostasis (PMID: 18406322). Conversely, the upregulation of LPIN1 is associated with metabolic syndromes, including obesity and type 2 diabetes, making its mRNA a potential target for knockdown therapies (PMID: 22451040). Experimental approaches using antisense oligonucleotides (ASOs) or siRNA aim to modulate LPIN1 levels to improve insulin sensitivity and reduce hepatic steatosis. However, therapeutic development must navigate the narrow safety window to avoid inducing myopathy or systemic metabolic disruption.
