Overview

Phosphatidylcholine (PtdCho) biosynthesis refers to the cellular processes that generate phosphatidylcholine, a major glycerophospholipid component of eukaryotic cell membranes. PtdCho is essential for membrane structure, lipid metabolism, and signaling. Its synthesis occurs in nearly all mammalian cells and is also present in plants, yeast, and some bacteria. The two primary pathways are the CDP-Choline Pathway (Kennedy Pathway) and the Phosphatidylethanolamine Methylation Pathway. Key enzymes include choline kinase (CK/ChoK), CTP:phosphocholine cytidylyltransferase (CCT/CCTα/CCTβ), and CDP-cholinetransferase. Regulation occurs at multiple levels, including enzyme translocation and transcriptional regulation. Alterations or upregulation of this pathway are observed in cancer cells due to increased demand for membrane synthesis during rapid proliferation.
