Overview

Phosphatidylethanolamine-binding protein 4 (PEBP4) is a scaffold protein and a member of the PEBP family, which includes the well-known Raf kinase inhibitor protein (RKIP) (UniProt Q96S96) [1]. Unlike RKIP, PEBP4 typically functions as an oncogene and a potent anti-apoptotic factor by interacting with and modulating key signaling cascades, such as the Raf-1/MEK/ERK and PI3K/Akt pathways (PMID: 15611084) [2]. PEBP4 is frequently overexpressed in a wide range of human cancers, including breast, lung, and colorectal carcinomas, where its high expression correlates with poor prognosis and increased metastatic potential (PMID: 25670303) [3]. Its role in promoting resistance to apoptosis induced by chemotherapy or radiation makes it a significant therapeutic target (PMID: 24333725) [4]. Targeting PEBP4 mRNA via RNA interference (RNAi) has demonstrated the ability to restore sensitivity to standard treatments in preclinical models. While no clinical drugs are currently approved, PEBP4 remains a focus of research as both a therapeutic target and a diagnostic biomarker for aggressive disease.
