Phosphatidylinositol 3-kinase-Akt kinase pathway kinases comprise a set of enzymes central to the PI3K-Akt signal transduction pathway, which orchestrates cellular survival, proliferation, metabolism, migration, and angiogenesis in response to growth factors and extracellular signaling[1][2][3][4]. PI3K kinases activate the pathway by generating the second messenger PIP3, which recruits and activates Akt kinases (protein kinase B isoforms: Akt1, Akt2, Akt3). Activated Akt phosphorylates numerous downstream effectors that regulate cell cycle progression, metabolism, apoptosis resistance, and other essential processes[1][2][3]. Mutational dysregulation of these kinases is strongly implicated in cancer, metabolic syndromes, cardiovascular, and inflammatory diseases. Both PI3K and Akt are independently druggable targets, and therapeutic development continues for their selective and pan-inhibitors, especially in oncology due to the frequency of their aberrant activation or mutation[5]. Safety and efficacy concerns remain due to the ubiquity of this pathway in normal cell physiology and its role in metabolic regulation.
