Overview

Phosphatidylinositol 3-kinase beta (PI3Kβ) is a Class IA PI3K, a lipid kinase that phosphorylates phosphoinositides. It is a heterodimer composed of a catalytic subunit (p110β) and a regulatory subunit (p85, p55, or p50). PI3Kβ integrates signals from receptor tyrosine kinases (RTKs), G protein-coupled receptors (GPCRs), and small GTPases. It plays critical roles in various physiological processes, including spermatogenesis, macrophage function, osteoclast development, neutrophil function, and platelet activation. Dysregulation of PI3Kβ is linked to cancer, thrombosis, male infertility, and Fragile X syndrome. Selective inhibitors are under development as potential therapeutics.
