Overview

Phosphatidylinositol 3-kinase catalytic subunit type 3 (VPS34, PIK3C3) is a lipid kinase belonging to Class III PI3Ks. It catalyzes the phosphorylation of phosphatidylinositol (PI) to generate phosphatidylinositol 3-phosphate (PI3P), which is essential for membrane trafficking pathways, especially those regulating autophagy and endosomal sorting. VPS34 is ubiquitously expressed and plays a unique role compared to Class I and II PI3Ks. It is emerging as a therapeutic target in cancer and other diseases where autophagy, vesicle trafficking, and cellular metabolism are dysregulated.
