Phosphatidylinositol 3-kinase delta (PI3Kδ) is a lipid kinase and a member of the class I phosphoinositide 3-kinases. It functions as a heterodimer composed of a catalytic subunit (p110δ) and a regulatory subunit (typically p85α or p85β). PI3Kδ is highly expressed in leukocytes and plays a central role in immune cell signaling, particularly in B cells and T cells, mediating survival, migration, and activation. Dysregulation of PI3Kδ is associated with immunodeficiency syndromes (e.g., APDS) and hematological malignancies, making it a validated therapeutic target. Selective inhibitors, such as idelalisib, have been developed for the treatment of certain blood cancers.
