Overview

The Phosphatidylinositol 3-Kinase (PI3K) family is a group of intracellular enzymes that phosphorylate phosphoinositides, playing crucial roles in cell growth, proliferation, survival, and metabolism. They are divided into three classes (I, II, and III) based on structure and substrate specificity. Dysregulation of PI3K signaling is frequently observed in cancer, making it an attractive therapeutic target. Class I PI3Ks are the most studied due to their central role in cancer biology and immune regulation. Several small molecule inhibitors targeting specific isoforms have been developed.
