Overview

Phosphatidylinositol 3-kinase (PI3K) regulatory subunit gamma is a component of the PI3K enzyme complex, which plays a central role in cell signaling pathways that regulate growth, survival, proliferation, motility, and immune responses. It is part of class IB PI3Kγ, existing as heterodimers composed of a p110γ catalytic subunit paired with either a p101 or p87 (also known as p84) non-catalytic regulatory subunit. These non-catalytic subunits determine the specificity and regulation of PI3Kγ activity in response to upstream signals. PI3Kγ is primarily involved in linking GPCR activation to downstream production of phosphatidylinositol 3,4,5-trisphosphate (PIP₃), regulating leukocyte chemotaxis, modulating immune cell functions, and acting as a scaffold for protein kinase A (PKA) complexes. Dysregulation or mutation can contribute to inflammatory diseases or cancer progression.
