Overview

Phosphatidylinositol 4,5-bisphosphate 3-kinase beta (PI3Kβ) is an isoform of the class IA phosphoinositide 3-kinases (PI3Ks), a family of lipid kinases that play central roles in cellular signaling. PI3Kβ specifically catalyzes the phosphorylation of phosphatidylinositol-4,5-bisphosphate (PIP2) to generate phosphatidylinositol-3,4,5-trisphosphate (PIP3), a second messenger that regulates diverse cellular processes including growth, proliferation, survival, metabolism, and motility. It is a heterodimer composed of a regulatory subunit (often p85α or its variants) and a catalytic subunit (p110β). PI3Kβ has both kinase-dependent and kinase-independent functions, most notably in DNA replication and cell cycle progression in addition to its role in canonical PI3K/AKT/mTOR signaling.
