Overview

Phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit beta (PI3Kβ or p110β) is the catalytic subunit of class IA phosphoinositide 3-kinases (PI3Ks). These enzymes regulate cell growth, proliferation, differentiation, survival, and metabolism by phosphorylating phosphatidylinositols, generating PIP₃. PI3Kβ plays a critical role in the PI3K/AKT pathway and has unique nuclear functions, including DNA replication control, distinguishing it from other class I isoforms. Dysregulation of PI3Kβ is implicated in oncogenesis, making it a target for cancer therapy. PTEN antagonizes PI3Kβ activity. Several inhibitors targeting different isoforms have been developed for cancer therapy; some drugs such as copanlisib target multiple class I isoforms including β.
