Phosphatidylinositol‑4‑kinase beta (PI4KB) is a key enzyme involved in the phosphorylation of phosphatidylinositol (PI) to generate phosphatidylinositol 4-phosphate (PI4P), an essential lipid signaling molecule that regulates various cellular processes, particularly membrane trafficking and organelle identity. It is crucial for defining Golgi and trans-Golgi network membranes, regulating vesicular trafficking from the Golgi to plasma membrane and maintaining structural integrity of the Golgi complex. Many positive-sense single-stranded RNA viruses hijack human PI4KB to create specialized membranous webs enriched in PI4P, which serve as platforms for viral replication. Additionally, Plasmodium falciparum's homologous enzyme is targeted by antimalarial imidopyrazines.
