Overview

Phosphatidylinositol 4-kinase type 2 beta (PI4K2B) is a membrane-associated lipid kinase that catalyzes the phosphorylation of phosphatidylinositol (PI) at the D-4 position of the inositol ring, generating phosphatidylinositol 4-phosphate (PI4P)[4]. PI4P defines the identity of the Golgi and trans-Golgi network membranes, plays a role in vesicle and endosomal trafficking, and acts as a precursor for phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2), a critical mediator in numerous signaling processes[4]. PI4K2B, along with its isoform PI4K2A, is classified as a type II PI4K; both differ from type III PI4Ks in sequence, structure, and regulatory mechanisms. While PI4K2A is the major contributor to PI4P synthesis, PI4K2B is implicated in cellular trafficking, immune activation, and potentially in early T cell signaling. Dysfunction or altered activity of PI4K2B has been linked to cancers, neurodegenerative diseases, immune responses, and viral replication cycles[4]. As a target, its unique structure and membrane interactions offer opportunities for drug design but also pose safety risks due to its central role in basic cell physiology.
