Phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 beta (PIK3C2B) is a class II phosphoinositide 3-kinase that phosphorylates phosphatidylinositol and phosphatidylinositol-4-phosphate, contributing to the generation of phosphoinositides involved in signaling and membrane dynamics. PIK3C2B regulates critical pathways including cell proliferation, migration, and survival in diverse tissues. It contains a C2 domain that allows calcium-independent binding to phospholipids, targeting the enzyme to cellular membranes. PIK3C2B is broadly expressed in cytoplasm and nuclei of most tissues. This kinase has been implicated in various cancers (notably prostate, esophageal, and ovarian) and other diseases like myotubular myopathy, by promoting or modifying cell signaling pathways important for tumor progression, cell invasion, and disease modifier effects. Pharmacological or genetic inhibition of PIK3C2B has demonstrated disease-modifying potential in certain models, notably in myotubular myopathy, and can decrease cancer cell invasiveness and metastatic potential.
