Phosphatidylinositol-4-phosphate 5-kinase like 1 (PIP5KL1) is a protein-coding gene encoding a kinase-like molecule that lacks intrinsic lipid kinase activity. Instead, PIP5KL1 acts as a scaffold, associating with type I phosphatidylinositol 4-phosphate 5-kinases and localizing their activity within specific subcellular compartments. Through this scaffolding function, PIP5KL1 facilitates the generation of phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2), which is critical for actin nucleation, cell signaling, and recruitment of scaffold proteins for further signaling cascades or conversion to PI(3,4,5)P3. PIP5KL1 is not an active kinase and is not considered a direct receptor, enzyme, or conventional therapeutic target. It has only limited associations with disease, such as reported connections to multiple types of cataracts.
