Overview

Phosphatidylinositol-5-phosphate 4-kinase type 2 beta (PIP4K2B) is an enzyme that catalyzes the phosphorylation of phosphatidylinositol 5-phosphate (PI(5)P) to generate phosphatidylinositol 4,5-bisphosphate (PI(4,5)P₂), a critical lipid messenger involved in plasma membrane signaling, cytoskeletal regulation, and vesicular trafficking. PIP4K2B is part of the phosphatidylinositol-5-phosphate 4-kinase family and plays dual roles as a sensor and regulator, controlling homeostatic PI(4,5)P₂ synthesis by inhibiting PIP5K enzymes. It is distinguished by prominent nuclear localization, where it influences chromatin organization, nuclear envelope mechanics, and mechanotransduction. Its expression is upregulated in certain cancers and correlates with poor prognosis. PIP4K2B modulates signaling pathways such as PI3K/Akt and YAP and regulates insulin sensitivity. Although not currently the direct target of approved drugs, pharmacological inhibitors have revealed its importance in cellular mechanics and cancer cell motility.

Mechanism of action

Inhibition of PIP4K2B disrupts nuclear envelope tension, chromatin compaction, and cell motility, interfering with YAP signaling and transcriptional regulation. Inhibition indirectly modulates PI(4,5)P₂ levels and downstream PI3K signaling, including Akt phosphorylation.
