The phosphatidylinositol glycan anchor biosynthesis class U protein (PIGU) is a transmembrane enzyme and an essential component of the glycosylphosphatidylinositol-anchor (GPI-anchor) transamidase complex, mediating the post-translational attachment of GPI anchors to proteins in the endoplasmic reticulum[1][2][3][7]. PIGU helps recognize or bind the lipid portion of the GPI-anchor and organizes other subunits within the membrane, which is crucial for GPI-anchored protein biosynthesis. Mutations in PIGU can lead to neurodevelopmental disorders, and its overexpression has been linked to cancer progression through immune evasion and increased NF-κB pathway activation[3][5][6][7].
