Phosphatidylinositol glycan anchor biosynthesis class Y protein (PIGY) is a subunit of the glycosylphosphatidylinositol-N-acetylglucosaminyltransferase (GPI-GnT) complex, which catalyzes the initial step in the biosynthesis of glycosylphosphatidylinositol (GPI) anchors in the endoplasmic reticulum[1][3]. GPI anchors are glycolipid modifications used to covalently anchor a wide range of proteins to the cell membrane, impacting cellular functions such as cell-cell interactions and protein localization[1][3]. PIGY may act by regulating the catalytic subunit PIGA in this multicomponent enzyme complex[3]. Mutations in PIGY are associated with rare congenital disorders, specifically hyperphosphatasia with impaired intellectual development syndrome 6 (HPMRS6)[3]. No direct drugs are known to target PIGY, and it is not typically considered a standalone therapeutic target.
