Phosphatidylserine lipase ABHD16A (ABHD16A) is a serine hydrolase enzyme belonging to the alpha/beta hydrolase domain family. It is primarily responsible for the hydrolysis of phosphatidylserine to generate lysophosphatidylserine, a bioactive lipid involved in the regulation of immune signaling and inflammation[5][6]. ABHD16A is highly expressed in diverse tissues, including the brain and immune cells, and plays critical roles in lipid metabolism, neurodevelopment, immunomodulation, and mitochondrial dynamics[1][2][3][6]. Disruption or abnormal function of ABHD16A is linked to neurodegenerative diseases (notably hereditary spastic paraplegia), inflammatory conditions, cancer, and metabolic disorders. While no approved drugs target ABHD16A directly, its pathway is considered a promising target for future therapeutic development in immunological, neurological, and metabolic diseases due to its pivotal role in lysophosphatidylserine metabolism and cellular signaling[2][3][6].
Inhibition of ABHD16A reduces hydrolysis of phosphatidylserine, thus altering levels of lysophosphatidylserine and downstream immune signaling. Regulation of neuroinflammation or immune responses through lyso-PS pathway moderation.
