01

Overview

Phosphatidylserine lipase ABHD16A (ABHD16A) is a serine hydrolase enzyme belonging to the alpha/beta hydrolase domain family. It is primarily responsible for the hydrolysis of phosphatidylserine to generate lysophosphatidylserine, a bioactive lipid involved in the regulation of immune signaling and inflammation[5][6]. ABHD16A is highly expressed in diverse tissues, including the brain and immune cells, and plays critical roles in lipid metabolism, neurodevelopment, immunomodulation, and mitochondrial dynamics[1][2][3][6]. Disruption or abnormal function of ABHD16A is linked to neurodegenerative diseases (notably hereditary spastic paraplegia), inflammatory conditions, cancer, and metabolic disorders. While no approved drugs target ABHD16A directly, its pathway is considered a promising target for future therapeutic development in immunological, neurological, and metabolic diseases due to its pivotal role in lysophosphatidylserine metabolism and cellular signaling[2][3][6].

Mechanism of action

Inhibition of ABHD16A reduces hydrolysis of phosphatidylserine, thus altering levels of lysophosphatidylserine and downstream immune signaling. Regulation of neuroinflammation or immune responses through lyso-PS pathway moderation.

03

Biological functions

Lipid metabolism: hydrolyzes phosphatidylserine (PS) to generate lysophosphatidylserine (LPS)Immunomodulation: regulates immune signaling, especially via lysophosphatidylserine (lyso-PS) and cytokine production in macrophagesRegulation of mitochondrial fission and fusion, affecting intracellular dynamicsDepalmitoylation of IFITM proteins, impacting antiviral protein traffickingRegulation of acylglycerol metabolism and possibly prostaglandin-glycerol hydrolysisNeural lipid homeostasis and neurodevelopment
04

Disease associations

Neurodegenerative diseases (including hereditary spastic paraplegia)InflammationImmunological disorders (e.g. Kawasaki disease)Cancer progression (ABHD16A-related microRNA implicated in hypoxia pathways)Metabolic diseases (altered lipid metabolism)
05

Safety considerations

Disruption of ABHD16A function may lead to immune dysregulation, neurodevelopmental defects, or altered lipid metabolism, which could result in neurodegenerative or inflammatory diseasesTherapeutic challenges include specificity (broad family of ABHD enzymes), tissue-specific effects (neurological vs. immunological roles), and potential metabolic disturbances
