Phosphodiesterases (PDEs) are a superfamily of enzymes that catalyze the hydrolysis of phosphodiester bonds, primarily in cyclic nucleotides such as cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP). By breaking down these second messengers, PDEs play a central role in regulating intracellular signaling pathways that control numerous physiological processes. There are 11 distinct mammalian PDE gene families, named PDE1 through PDE11. Each family can have multiple subtypes and isoforms due to alternative splicing; over 60 isoforms have been identified in humans. Dysregulation or genetic mutations affecting specific phosphodiesterase genes have been linked with various pathologies and represent both diagnostic markers and therapeutic opportunities.
Hydrolyzes phosphodiester bonds in cyclic nucleotides (cAMP and cGMP), reducing their intracellular concentrations and modulating downstream signaling pathways.
