Phosphodiesterase 2A (PDE2A) is a dual-substrate enzyme that hydrolyzes both cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), playing a key role in the regulation of intracellular signaling pathways mediated by these second messengers. PDE2A belongs to the phosphodiesterase family, specifically the PDE2 subfamily, and is encoded by the PDE2A gene in humans. PDE2A is allosterically activated by cGMP binding, influencing cAMP hydrolysis. It is involved in various cellular functions, including PKA activation and natriuretic peptide signaling. Aberrant PDE2A expression or activity is implicated in several diseases, including intellectual disability syndromes, movement disorders, cancer progression, hypertension and heart failure, making it a potential therapeutic target.
