Overview

Phosphodiesterase 3B (PDE3B) is an enzyme that belongs to the PDE3 family. It regulates the intracellular concentrations of cyclic nucleotides, cAMP and cGMP, by catalyzing their hydrolysis. PDE3B is highly expressed in tissues involved in energy metabolism such as hepatocytes, brown and white adipocytes, and pancreatic β cells. It regulates key metabolic processes including lipolysis, glucose uptake, and insulin secretion, making it a potential therapeutic target for metabolic disorders like obesity and type II diabetes mellitus. Its involvement in cardiovascular function further broadens its clinical relevance. Alterations in PDE3B have been linked to conditions like primary aldosteronism and vitamin D hydroxylation-deficient rickets type 1b. circRNA forms of this gene have been implicated in cancer progression.

Mechanism of action

PDE3B inhibitors increase intracellular cAMP and cGMP levels by preventing their degradation, leading to various downstream effects depending on the tissue.
